收到區域法院信件,如對信件內容作出犯對需於21天內以書面回覆,如超過21天後才提出是否有問題?

Mostly yes.
As a legal assumption, if you have not responded within the deadline, you are assumed to give up your right to contest. Although the Court may accept late response as a matter of discretion, still late response is a trouble by itself.
Note - A request to extend deadline is a response.
