Your testicles don't "fill". They are solid organs, and won't get bigger from abstaining from solo sex. Semen is produced in the prostate gland. Holding off masturbation may well result in a wet dream -- nocturnal emission of semen. Most guys don't actually have a "dream" they just wake up with messy underwear or sheets.

Unless you have see-through testicles, you won't notice any difference in the amount of ejaculate stored there, nor size.
