If I die even once, let alone more than that, will it affect the overall ending of the pacifist run?

Nope. Someone may comment on it, if you get to a certain place with no deaths, but it's a cosmetic difference at the most. It won't affect a thing otherwise.

Yes and no. Yes there will be very few dialogue changes based on the number of deaths you have. No a pacifist run will not expire if you die too many times.
