My husband is ~ 260 lbs. He wants to repair a rip in our pool screen that would require him to go on the roof. Could he fall through?

Yes.
Update: Yes.

Large people that are used to being on the ground all the time need to do themselves a favor and stay on the ground. Just standing at an angle will wear him out. If he loses his balance and falls off, injury is very likely. If something is mechanically unsound (rotten) he could fall through. A broken back is a bad injury. Hire somebody!

Tell your Husband to hire a roof Ladder. That will not damage the roof and the safest way to work on a roof.

The biggest danger is that he is not athletic enough to move easily and not flexible enough to avoid injury if he falls off. Tell him to hire somebody. It's cheaper than an ambulance.

In the US, Google your city or county + snow load requirement. Everywhere has one, even places that never get snow. You should get a local government website that should tell you how many pounds per square foot your roof is required to hold, if it's up to code.

if the roof is rotted he could fall through

if he has balance problems he could just fall off

It would be prudent to use crawl boards or ladders. Your roof should be strong enough to take the weight, but it depends on its type and construction.
