a failure, defect, or deficiency in conduct, condition, thought, ability, etc.
This meaning referring to conduct, condition, thought, ability is similar yet different from the meaning of weakness in 2 above which refers to a person's quality such as their character..

如果你查字典，你會發現二字是同義詞 synonym。
一般來說可以互用，但如果你希望細分，其實也可以找到不同之處。

shortcoming 是指缺點，
weakness 是指弱點。

Yahoo!字典：
Most people saw her sensitivity as a sign of weakness.
許多人把她的敏感看作是懦弱的表現。

這裏不能用 shortcoming。
