Boltbus.com provides the best service for the price. However, for complete schedule flexibility look at us.megabus.com and peterpanbus.com and also greyhound.com. The trip takes under 5 hours.For a faster ride there is Amtrak. Schedule ticket two weeks or more in advance for best fares.
Reservations are required on all of the above, but greyhound and amtrak do sell empty seats on the day of departure for a significant markup.

It depends on what bus you take. I took both grey hound and mega bus from Boston to NYC and both were at least an hr over schedule due to traffic when getting into NYC. However Mega bus was cheaper, but they drop you off on a side street in NYC because their bus terminal can't fit their buses.
I would say about 4-5hrs almost 5 and a half going to NYC and that includes the traffic you will hit once you get to NYC.
About 5 going out because it is a little easier getting out of the city than it is getting out.
Mind you if you take mega bus from boston you pick it up at the south station bus station, and it drops you off on a side street, and when taking it back to boston check your ticket because it picks up in a different spot from where it dropped off. Plus you MUST have the ticket for the correct bus time, you can not get on an earlier bus unless they have room or a later bus unless you miss your scheduled bus.
Grey hound I have taken round trip to NYC for $100 and the earliest bus they had was 7am, megabus leaves at 6am. The earliest they had was the 7am one. Grey hound is pretty good though about as long as your ticket is for the day you wanna get on the bus you should be all set.
Keep in mind again though megabus you gotta line up wicked early because the line at south station can go as far back as the ticket booths. It is crazy especially the people who take the last one back to NYC.

Greyhound is 4 hours and twenty minutes. if you are thinking of taking Greyhound my advice to you would be to book the ticket online 3 to 4 weeks before you go. That's when the tickets are the cheapest (as cheap as $11 each way).
