When air is pumped into the tyre, the tyre contains more number of air molecules than before. Since air molecules inside the tyre are moving randomly in all directions, the number of molecules striking the inner surface of the tyre increases.

Because pressure of a gas is due to the force given by molecules bombarding a surface, the increase in number of molecules striking the tyre surface leads to an increase of pressure.
