my sister and my sister's husband both are American citizens. My sister is unable to work. Can my sister file petition for me????

ACtually, depend on how your sister unable to work.....She may not be able to file your F-4 anyway.
IFshe is an able body person, she cannot use normal sponsorship form to sponsor you, she must be the one on the form to sponsor you.
Unless she is unable to work due to long term illness, mental illness and disability, she can then use secondary sponsor to sponsor you. Or, simply, if your sister is over the retire ages, she can also use other people to sponsor you.

if your sister sponsor you and your family, YES, he can.

It depends on what you mean.
1. In term of immigration eligibility, no. In a simple way, your sister-in-law can't file a I-130 to apply you to the United States. You sister must be the one.
2. In term of financial support (I-864), yes. In a simple way, use I-864A.
