Actually it's not that difficult as u think.
It comes from the Principle of relativity.
Here's just some simple points.
The Relativity states that the velocity of an object can't exceed the speed of light, so we now that the kinetic energy is not that 1/2mv^2.
By just using the equation of kinetic energy stated by Einstein,
and subs. v=0, and then we get E=mc^2

Einstein published his Theory of Special Relativity(特殊相對論,或名狹義相對論) in 1905. The equation E=mc2 was published in a short paper later in the year as a supplement to the Special Relativity Theory. This short paper (original version is in German, but English translation is available) titled "Does the inertia of a Body Depends on its Energy Content" can be read on the following link:

In that paper, Einstien derived this famous equation.
