首先, 就算中文你也不會問: 你能不能講廣東話? (can you speak ....? 不對)
所以, 你應問: do you speak Cantonese? (你識唔識講廣東話?)

Can you speak cantonese ? It is the right one.
We do not use speak "in" . But we can use speak to, of, out/up, for.
for example: Can you speak English? He is speaking to me.
Are you speaking of me? You have to speak up loud. He spoke for two hours already.
