How to transfer bacterial colony from a plate to another?

If I have a colony of bacteria growing on a plate and I want to transfer it aseptically to another plate. What should I do? Do I transfer some of it with a needle to a broth, mix it well and streak this broth onto another plate?

If you don't have access to disposible loops, you can make one from wire, use the tip of a sharp pencil to form the loop, that will be about the size of a 10 uL. You will need to flame the loop, give it a minute to cool then transfer the colony to the desired plate. Use either a 3 or 4 way streak for Isolation.
You can always use it like a needle too, just be careful not to plough up the agar.

Used to have to do it that way til we got sterile loops.

subbing it out to another plate would be ideal...i would'nt transfer it to a broth you risk chance of contamination...use a disposable loop!! and make sure you streak it as to isolate the colony just in case you have more than one colony overlapping one another!

You can do that, or you can use a loop or straight inoculating wire to pick up bacteria from an isolated colony on the plate and streak it onto another plate. Using aseptic technique of course.
