I've learned a lot about Sickle-cell anemia (disease) in my Biochemistry lectures, I'll put it in an easy way, hope this can help you.
People with sickle cell disease will have Red Blood Cell with the shapes like sickle (鐮刀形狀). Changing the shape of cell membrane, it damages the Red Blood Cell membrane, and it will get stuck in the blood vessels, decreasing the oxygen binding efficiency. Therefore, people will deprive of oxygen, and result ischemia.
People normally die at age of 40. People in Africa are more prone to have this, and if people have heterozygous allele (one is sickle cell, the other is normal), that is, they're a carrier. This actually guards them against malaria, which helps a bit in countries with low quality water.
